Interferon (IFN)-alpha has been used clinically to treat various malignancies. IFN is also known to induce a number of pharmacological effects. However, the effect of IFN on natural killer (NK) cell activity is not known. The immunological function of various peripheral blood lymphocytes in uremic patients with various types of dialysis was examined before and after IFN therapy. A rapid recovery of leukocyte count was observed after IFN therapy, and lymphocyte subsets were changed to normal. NK cell activity was markedly decreased in these patients, and this reduction was restored after IFN therapy. Lymphocytes from dialysis patients did not show significant cytotoxicity, whereas those from healthy subjects showed relatively high cytotoxicity. IFN reduced NK cell activity in healthy subjects, but not in dialysis patients. NK activity recovered to normal after IFN therapy in dialysis patients. These findings suggest that IFN therapy increases the ratio of active cytotoxic lymphocytes and increases the cytotoxicity of those lymphocytes in the circulation, as well as increasing lymphocyte number. These phenomena may explain the beneficial effects of IFN therapy in uremic patients.Molecular dynamics of lipid bilayers and proteins: a new method for simulation studies.
